چکیده

PURPOSE The expression of heat shock protein 47 (HSP47) was observed histologically to investigate the spatial and chronological effects of excimer laser photoablation. HSP47 expression after radial keratotomy (RK) was also investigated and compared with the effects after excimer laser photoablation. METHOD Twenty-eight male rabbits were used. The rabbits were divided with two groups and treated with either excimer laser photoablation or four radial incisions to simulate corneal refractive surgery. The chronological and spatial changes in the expression of HSP47 were observed immunohistochemically. RESULTS In eyes that underwent excimer laser photoablation, HSP47 was detected in the basal layer of the epithelial cells and in the superficial stromal layer 3 days after surgery. After 5 and 7 days, HSP47 expression extended to the deep layer of the stroma and to the endothelial cells. After 14 days, HSP47 was detected only in the deep layer of the stroma and in the endothelial cells. After 28 days, HSP47 expression was reduced. In eyes that underwent RK, HSP47 was detected in the basal layer of the epithelial cells and in the stroma surrounding the wound 1 day after surgery. After 3 and 7 days, HSP47 expression did not expand further. After 28 days, HSP47 expression diminished. CONCLUSIONS Excimer laser photoablation affects the whole layer of the cornea, and may be caused by the shock wave that occurs as a result of photoablation. In addition, interaction among the keratocytes may propagate the stress-induced response to the whole layer of the cornea. With RK, the wound is smaller and deeper. HSP47 expression occurs earlier, but is limited to the area surrounding the wound.

Effect of Thermal Preconditioning Before Excimer Laser Photoablation

The purposes of this study were to assess the expression patterns of heat shock proteins (Hsps), after eyeball heating or cooling, and to elucidate their relationships with corneal wound healing and intraocular complications after excimer laser treatment. Experimental mice were grouped into three according to local pretreatment type: heating, cooling, and control groups. The preconditioning was...

Gas chromatographic and mass spectroscopic analysis of excimer and erbium: yttrium aluminum garnet laser-ablated human cornea.

The photoablative products from human cadaver corneas treated with lasers were investigated using gas chromatography and mass spectroscopy. The photoablations were done separately with an excimer laser (193 nm) and with an erbium: yttrium aluminum garnet (Er:YAG) laser (2.94 microns). More than 20 different types of molecules with a molecular weight of 40-400 mass units could be identified, mos...

Corneal aldehyde dehydrogenase and glutathione S-transferase activity after excimer laser keratectomy in guinea pigs.

BACKGROUND The free radical balance of the eye may be changed by excimer laser keratectomy. Previous studies have demonstrated that excimer laser keratectomy increases the corneal temperature, decreases the superoxide dismutase activity of the aqueous, and induces lipid peroxidation in the superficial corneal stroma. Aldehyde dehydrogenase (ALDH) and glutathione S-transferase (GST) are known to...

Analysis of glycosaminoglycans in rabbit cornea after excimer laser keratectomy.

BACKGROUND/AIMS The biochemical basis for the development of subepithelial opacity of the cornea after excimer laser keratectomy has yet to be fully defined. The aim of this study was to evaluate the alterations of glycosaminoglycans (GAGs) after excimer laser keratectomy. METHODS Rabbit corneas were harvested on days 5, 10, 20, and 30 after excimer laser photoablation. The amount of main dis...

EVects of amniotic membrane on epithelial wound healing and stromal remodelling after excimer laser keratectomy in rabbit cornea

Aims—To investigate if the amniotic membrane (AM) promotes epithelial migration while inhibiting stromal remodelling associated with corneal haze after excimer laser keratectomy. Methods—A wound 150 μm in depth and 6.0 mm in diameter was produced in 40 rabbits using an excimer laser. One eye was randomly chosen to be covered by the AM while the other eye served as a control. Epithelial wound he...
